Our approach
Leyden Labs is built on a simple premise: To stop respiratory viral infections, we need to protect at the gate where viruses invade the body. We are creating a new class of medicines called Mucosal Protection Products that will introduce the world to completely new ways to fight respiratory infections, placing protection in the palm of the hand, to use when and where needed.
Mucosal Protection Products are Designed for Breadth
Unlike existing solutions, Mucosal Protection Products are designed to block viruses before they take hold. With these product candidates, we aim to deliver unprecedented breadth of protection against entire viral families, providing coverage against virus strains and variants that already cause illness, and those that might in the future.

Delivery of Mucosal Protection Products to the airway is made possible by our world-class molecules, expertise in formulation science, and targeted airway delivery capabilities.



Product Pipeline
We are advancing two distinct mucosal approaches to protect against respiratory viruses: broadly protective antibodies and innate immune activators. Both mucosal approaches aim to block viral replication at the gate of entry in order to prevent illness, including progression to the lungs and severe disease.
Broadly Protective Antibodies
Our broadly protective antibodies provide, for the first time, the opportunity to deliver antibodies intranasally and provide immediate neutralization of viruses at the mucosal surface. These antibodies are uniquely broad in their protective activity because they target the most conserved mechanisms of viral infection. As a result, these antibodies will continue protecting even if and when a virus changes over time.
Our lead antibody program, PanFlu, is designed to deliver first-in-class protection against all influenza viruses by using intranasal administration of CR9114 – the most broadly active flu antibody identified to date.
Innate Immune Activators
Our innate immune activators, for the first time, enable delivery of oligonucleotides directly to the airway to stimulate natural antiviral defenses. With this approach, the goal is to prime the immune system for an appropriate and effective defense against viral pathogens. Innate immune activators thus provide a complementary approach to our broadly protective antibodies—providing two distinct ways to defend against respiratory viral infections.
Our lead program delivers a RIG-I agonist called RIG-101 directly to the airway. Based on its mechanism of action, RIG-101 is expected to prevent infections caused by a broad range of RNA viruses including human rhinovirus, respiratory syncytial virus, influenza, and coronavirus SARS-CoV-2.
RIG-101 is delivered to the upper airway using our proprietary NEED™ Delivery Platform—a non-LNP nanoemulsion delivery platform that has been optimized for intranasal delivery with excellent local tolerability characteristics. We are initially evaluating RIG-101 for its ability to prevent asthma exacerbations that are triggered by infection with human rhinovirus.
